When you call, simply say: "Hi, my name is _____________. I submitted an application/resume _______ weeks ago. Can you give me an update on the status of the job opening?" You can't really ask for an interview. At least, not that I have ever known. The employer will call you if they want to set one up.

"Hi my name is xxx i recently filled out an application form online for a (insert position), and was wondering if you have had chance to look over it yet?" Wait 2 -3 days after you submit your application and then call. EDIT With places like walmart, target etc. They receive hundreds of applications and its a very slim chance you will get a reply back. I would print out a bunch of resumes, and walk into places ask for jobs and hand out your resume. Try smaller shops, they are more likley to interview you than massive chains.

You call and ask to speak to the person who does the hiring. (9 times out of 10 - they ain't hiring) Tell them you dropped off your application and would like an interview. Ask questions like when are you interviewing - what jobs are open - and before you even ask how much the pay is, or when are the vacations - you let them know that you're willing to work any shift (if you are) that you want to start at the lowest level and work your way up.

call and ask if you can speak with the human resources department regarding your job application. ask for a hiring manager if they dont have an HR department. let them know that you visited on such and such day and filled out an application for whatever position. ask them if the position is available or if they have any other positions available. if the answer is yes, tell them you are very interested in employment with their company. ask them if you can schedule a time to discuss employment opportunities.
